Fangfang Bai is a Senior Scientist in the department of Data & Genome Science at Merck Research Laboratory Cambridge. She joined Merck since December 2019. She is leading the efforts on utilizing CRISPR screening and iPSC-derived human microglial-like cells to identify new therapeutic targets for neurodegenerative diseases. She has established the CRISPR screening platform and studied the functional consequences of genetic perturbations in iPSC-derived human microglial-like cells. Fangfang earned her PhD at Ocean University of China. I am currently an Associate Research Scientist in the Department of Neurology at Columbia University, New York. I am funded by an Early Career Investigator Innovation Award from the William N. and Bernice E. Bumpus Foundation. The focus of my research encompasses two overarching research questions: 1) Why and how do dopaminergic neurons selectively degenerate in Parkinson’s disease (PD)? and 2) What is the function of the alpha-synuclein protein, in which location, and how does too much endogenous alpha-synuclein contribute to PD? To begin to answer these research questions I use patient-derived induced pluripotent stem cell (iPSC) lines carrying pathogenic mutations in the alpha-synuclein gene SNCA. I directly differentiate these iPSC lines carrying endogenous levels of physiological alpha-synuclein into midbrain dopaminergic neurons and identify screenable functional phenotypes against healthy controls and/or Cripsr-Cas9 gene-edited isogenic controls. My current work is investigating the function of alpha-synuclein at the mitochondrial associated ER membranes (MAMs). These MAMs are transient, functional domains that regulate lipid homeostasis, and I pose that alpha-synuclein mediated modulation of MAM domains and regulation of lipid dyshomeostasis may contribute to PD neurodegeneration, and that the mechanism of cell death may be non-cell autonomous.
