Abstract

Purpose

Nanosecond pulsed electric fields (nsPEFs), a biophysical signal, is intimately involved in regulation of phenotypes and differentiation of the cells. In previous study, we found that nsPEFs dedifferentiated chondrocytes via activation of Wnt signaling pathway [1] while enhanced chondrogenic differentiation of MSCs. In current study, we optimized key parameters of nsPEFs (field intensity and duration) and explored mechanisms behind.

Methods and Materials

Primary Porcine bone marrow-derived stem cells (bMSCs) were subjected to nsPEFs with 16 parameters (duration: 10ns, 60ns, 100ns, 300ns) plus (: 5kV/cm, 10kV/cm, 20kV/cm, 30kV/cm). Gene expression of collagen (COL) I/II/X, aggrecan (AGG) and SRY-related high mobility group-box gene 9 (Sox9) were evaluated at day 14 after bMSCs were subjected to nsPEFs. Protein expressions of phosphorylation-JNK and phosphorylation-CREB were tested with western blotting at 0.5 hour.

Results

Both 10 kV/cm at 100ns and 20 kV/cm at 10ns of nsPEFs significantly increased gene expression of COL II (16.46 folds,p<0.01; 10.27 folds, p<0.01) and SOX9 (14.95 folds, p<0.01; 8.79 folds, p<0.05, Fig.1A) at day 14, and upregulated the glycosaminoglycan (GAG) production at day 14 respectively (1.49 folds, p<0.05;1.67 folds, p<0.01, Fig.1B and 1C). Expression of phosphorylation-JNK protein and phosphorylation-CREB protein were increased (Fig.2).

Conclusion

Our results demonstrated that nsPEFs significantly enhanced chondrogenic differentiation of MSCs with dose-dependent effects, possibly through mediation of JNK-CREB signaling pathway which remains to be confirmed. 18-icrs-figure1.jpg
