Session Description:

Correct functioning of the brain barriers, cerebral spinal fluid (CSF) dynamics and its perfusion through the brain parenchyma (termed as glymphatic system), are important for brain health and their disruption underlies numerous neurodegenerative disorders. While our knowledge of the BBB is expanding, as is our appreciation of the importance of CSF in communicating between the brain parenchyma and the periphery via CNS-draining lymphatics, our understanding of the blood-meningeal and blood-choroid plexus-CSF barriers and their contribution to glymphatic flow remain limited. Understanding the access route for macromolecules, such as antibodies, from the periphery to the CSF/CNS is crucially important for delivery of therapeutics. Moreover, understanding how meningeal lymphatics, the BBB, the blood-meningeal barrier, and the blood-choroid plexus barrier interact and communicate, and how failure of one barrier might trigger dysfunction of CSF production, dynamics and glymphatic flow, and waste clearance are critically important to design better drugs for preserving CNS fluid homeostasis and sustaining brain health through aging. In this session we will discuss interactions among different barriers from four different angles, focusing on meningeal lymphatic vessels, glymphatic flow, lymphatic clearance BBB function and the role of brain resident macrophages in regulation of brain barriers.
